The department was formed in July 2012 through the merger of the Department of Cell & Developmental Biology and the Department of Cell & Molecular Physiology. The new department brings together two strong traditions of excellence in basic science and translational research, and in pre- and post- doctoral training, including undergraduate, graduate and medical education.

The lab of Dr. Natasha Snider studies the biology and pathophysiology of the intermediate filament (IF) cytoskeleton. Specifically, we are interested in therapeutic approaches for rare genetic diseases caused by mutations in IFs or IF-regulatory proteins. Our main goal is to prevent or alleviate the misfolding and aggregation of IF proteins that is the hallmark of these diseases. We are seeking a highly motivated postdoctoral fellow to study mechanisms of IF proteostasis and pharmacological approaches for intervention.

The project will involve development of high throughput screening (HTS) assays using primary dorsal root ganglion neurons and astrocytes from rodent models of Giant Axonal Neuropathy and Alexander Disease and validation of hits. The research also entails the use and further development of patient- derived motor neurons, astrocytes, and brain organoids of these two diseases using induced pluripotent stem cells and CRISPR/CAS9 genome editing. The work will be carried out in close collaboration with the Human Pluripotent Stem Cell Core (HPSCC) stem cell core facility at UNC.

The position will require: working with mouse and rat models of Giant Axonal Neuropathy and Alexander Disease; isolation of primary astrocytes and neurons; cell culture; pharmacological studies using FDA approved libraries, kinase inhibitors, and novel intermediate filament targeting compounds; developing biochemical and cellular assays for intermediate filaments; assaying filament properties using confocal and electron microscopy on fixed cells/tissues and in vitro assembled proteins.
